I figure we could follow a Teen Titan style formula. More funny but enough serious to balance it out. If anything we want to captivate the reader in a healthy marriage of those elements.

When I think PO I think cooky. Like you said, chibi chracters invite humor. The RPG is the same way.

Originally PO was thought up to be comical. We expected some seriousness involved and we have gotten that on a much larger scale.

Originally there was a running theme for Shino where things just had to fall on his head.

Okay. So I'll come to the conclusion based on what's here already:
Humorous, but yes, we will have our heads screwed on right for serious stuff. Origins can still be serious, but don't expect for the origin to be explained in the comic. ASSUMING the ongoing is something I(or we) can carry on on at LEAST a two/three issue a year basis(that sounds do-able...1 issue for every 3 months gives us about 4 a year or if we go the path of two or three a year, that's a little more time per comic), then there will be opportunities for origins to be shown.
Even though there are "bad" and "good" character in PO, I will take the liberties of creating new "specifically for the purpose of being killed" PO's/Enemies.

Pilot Issue idea:
Still working. This pilot will just introduce a new problem to Miniopia-

Pilot Issue Step-by-step(keep in mind, I don't know if there's a SPECIFIC way that EVERYONE in PO met. If there is a dissaproval/conflict of origins, let me know):
I want to start off with a flashback of Doot and Shino training under their master(forgot the name). The flashback will cut off and on between a chase (present) where Doot is trying to escape Poo-Ting's ninja's in a forest. The PO's that will be introduced in this scene (keep in mind that this is PRESENT PO date. ALL PO's are existing at this moment) are Shino, Doot, Cobalt Katze and Chibi Knight(brief appearance).
From there, it will move to Aind', Sakura, and Pickle at the Kilted Wench with some problems of their own( more ninjas). Introduction of Poo Ting General "Graizer". (I have him designed. I'll post it all up later when I have time). This is where Graizer will do the classic "explain what Poo-Ting's plans are" thing. Just to eat up more dead trees and stuff. (Aindriahhn, PRG, Sakura((Pickle)), and Sciler are in this scene)
Cut to the PO tournament arena- invaded by Poo-Ting's ninjas and General Achsis(axis). BWS-1, Gryph, Unastrike, Reverend, and a few other small PO cameos in this one.
Comic will end with Poo-Ting ripping a whole in time-and-space to release an AU creature upon Miniopia : D ( I WILL do something with the AU ideas >D)
