Lupin is looking at the map on that fateful night because he has a hunch (a very good one) that Harry and the others will be sneaking down to Hagrid's to see him before Buckbeak's execution. When the three of them leave Hagrids with Scabbers/Peter now in tow he sees his name on the map, and sees Black drag Scabbers/Peter into the tree and to the Shrieking Shack. Lupin rushes out of his office to confirm what the map is telling him and the rest is history.
